How to Use

Dosage

Your doctor will tell you the correct dose, which may change based on your blood tests. Adults usually start with 30 mg (one tablet) once or twice a day, depending on your condition.

Method

Take the tablet by mouth with or right after food. Swallow the tablet whole. Do not chew, crush, or divide it.

Important

Your doctor will determine your maximum daily dose based on your needs. For children (3-18 years), the maximum is 0.20 mg per kg of body weight per day.

Possible Side Effects

COMMON
  • Nausea and vomiting. These are usually mild and go away quickly.
  • Dizziness
  • Numbness or tingling sensations (paresthesia)
  • Loss or decreased appetite
  • Muscle pain
  • Weakness
  • Rash
  • Decreased testosterone levels
  • High potassium levels in the blood (hyperkalemia)
  • Allergic reactions
  • Headache
  • Seizures
  • Low blood pressure (hypotension)
  • Upper respiratory tract infection
  • Difficulty breathing (dyspnea)
  • Cough
  • Digestive problems (dyspepsia)
  • Diarrhea
  • Abdominal pain, upper abdominal pain
  • Constipation
  • Muscle spasms
  • Back pain
  • Low calcium levels in the blood (hypocalcemia)
RARE
  • Hives (urticaria)
  • Swelling of the face, lips, mouth, tongue, or throat, which may cause difficulty swallowing or breathing (angioedema)
  • Unusually fast or strong heartbeat, which can be due to very low calcium levels in the blood (QT prolongation and ventricular arrhythmia secondary to hypocalcemia)

Important Warnings

  • Do not take Sinafex if you are allergic to cinacalcet or any other ingredient in this medicine.
  • Do not take Sinafex if your blood calcium levels are too low. Your doctor will monitor your calcium.
  • Tell your doctor if you have ever had seizures (fits). The risk of seizures can be higher if you have had them before.
  • Tell your doctor if you have liver problems.
  • Tell your doctor if you have heart failure.

  • Contact your doctor immediately if you experience numbness or tingling around your mouth, muscle cramps or spasms, or seizures, confusion, or loss of consciousness. These could be signs of very low blood calcium.
  • Low calcium levels can affect your heart rhythm. Tell your doctor if your heart beats unusually fast or strong, or if you have a known heart rhythm problem.
  • Tell your doctor if you start or stop smoking while taking Sinafex, as this can affect how the medicine works.
  • Children under 18 with parathyroid cancer or primary hyperparathyroidism should not take Sinafex.
  • Contact your doctor immediately if you experience swelling of your face, lips, mouth, tongue, or throat, which can make it hard to swallow or breathe (a serious allergic reaction).
