How to Use

Dosage

For hereditary tyrosinemia type 1, the usual dose is 1 mg per kilogram of body weight per day, adjusted by your doctor. For AKU, the recommended dose is 10 mg once a day.

Method

Take the capsule by mouth, usually once a day. If you have trouble swallowing, you can open the capsule and mix its contents with a small amount of water, apple juice, applesauce, or a liquid nutritional product, right before taking it. Take with food if you started the treatment that way.

Important

For hereditary tyrosinemia type 1, the total daily dose is 1 mg per kilogram of body weight. For AKU, the total daily dose is 10 mg. Your doctor will set your specific dose.

Possible Side Effects

COMMON
  • Eye inflammation (red or irritated eyes), cloudy vision, sensitivity to light, eye pain (these are common in hereditary tyrosinemia type 1 patients due to high tyrosine levels).
  • Lower numbers of platelets (which help blood clot) and white blood cells (which fight infection), or a lack of certain white blood cells (these are common in hereditary tyrosinemia type 1 patients).
  • Eye irritation and eye pain (these are very common in alkaptonuria patients).
  • Bronchitis (inflammation of the airways), pneumonia (lung infection), itching, and rash (these are common in alkaptonuria patients).
RARE
  • Eyelid inflammation (blepharitis, this is less common in hereditary tyrosinemia type 1 patients).
  • Increased white blood cell count, itching, skin inflammation (exfoliative dermatitis), and rash (these are less common in hereditary tyrosinemia type 1 patients).

Important Warnings

  • Your eyes will be checked before and regularly during treatment. If your eyes become red or you have other eye problems, contact your doctor immediately, as this might mean your diet isn't working well.
  • You will have blood tests during treatment to check if the medicine is working and to watch for side effects that could change your blood values.
  • If you are taking this medicine for hereditary tyrosinemia type 1, your liver will be checked regularly because the disease affects the liver.
  • Tell your doctor or pharmacist about all other medicines you are taking, as Nitisinone Dipharma can affect how some other medicines work (e.g., for epilepsy or blood thinning).
  • It is not known if this medicine is safe during pregnancy or breastfeeding. Do not breastfeed while taking this medicine. Talk to your doctor if you are pregnant, planning to be, or become pregnant.
