How to Use

Dosage

Type 1 tyrosinemia: 1 mg per kg body weight daily, adjusted individually by your doctor. For patients under 20 kg, the daily dose is usually split into two. AKU: 10 mg once daily.

Method

Take the capsules by mouth once a day. If you have trouble swallowing, you can open the capsule and mix its contents with a small amount of water, apple juice, applesauce, or liquid nutritional product. If you start taking the medicine with food, continue to do so throughout the treatment.

Important

For hereditary tyrosinemia type 1, the maximum recommended daily dose is 1 mg per kg of body weight. For AKU, the maximum recommended daily dose is 10 mg.

  • Eye inflammation (conjunctivitis)
  • Clouding and inflammation of the cornea

  • Decreased number of platelets (thrombocytopenia)
  • Decreased number of white blood cells (leukopenia)
  • Lack of certain white blood cells (granulocytopenia)
  • Eye irritation (keratopathy)
  • Bronchitis
  • Pneumonia
  • Itching (pruritus)
  • Skin rash
RARE
  • Eyelid inflammation (blepharitis)
  • Increased number of white blood cells (leukocytosis)
  • Skin inflammation (exfoliative dermatitis)

Important Warnings

  • Do not take if you are allergic to nitisinone or any other ingredient in this medicine.
  • Do not breastfeed while taking this medicine.
  • Have your eyes checked regularly before and during treatment. If your eyes become red or you notice other eye symptoms, contact your doctor immediately, as this may indicate an issue with your diet.
  • You will have blood tests during treatment to check if the medicine is working and to monitor for side effects related to blood values.
  • If you are taking this medicine for hereditary tyrosinemia type 1, your liver will be checked regularly because the disease affects the liver.

  • Tell your doctor about all other medicines you are taking, as Nitisinone Dipharma can affect how other medicines work, such as those for epilepsy (e.g., phenytoin) and blood thinning (e.g., warfarin).
  • Contact your doctor if you are planning to become pregnant or if you become pregnant while taking this medicine.
  • If you experience vision-related side effects, do not drive or use machines until your vision returns to normal.
