How to Use

Dosage

Take one 10 mg tablet once a day.

Method

Swallow the tablet whole with a glass of water. Do not chew or break it. You can take it with or without food, but it's best to take it at the same time each day.

Important

The maximum recommended dose is 10 mg per day.

Possible Side Effects

COMMON
  • Anemia (low red blood cell count) or low hemoglobin levels
  • Headache
  • Bronchitis (inflammation of the airways)
  • Nasopharyngitis (inflammation of the nose and throat)
  • Swelling, especially in the ankles and feet
  • Pharyngitis (sore throat)
  • Flu-like illness
  • Urinary tract infection (bladder infection)
  • Low blood pressure
  • Nasal congestion
  • Increased liver enzyme levels
  • Leukopenia (low white blood cell count)
  • Thrombocytopenia (low platelet count)
  • Flushing (skin redness)
  • Increased uterine bleeding
  • Upper respiratory tract infections (e.g., infected nose, sinuses or throat) (in children)
  • Gastroenteritis (inflamed stomach and intestines) (in children)
  • Rhinitis (itchy, runny or stuffy nose) (in children)
RARE
  • Allergic reactions (swelling around eyes, face, lips, tongue or throat, itching and/or rash)

Important Warnings

  • Do not take if you are allergic to macitentan or any other ingredient in this medicine.
  • Do not take if you are pregnant, planning to get pregnant, or could become pregnant and are not using reliable birth control.
  • Do not take if you are breastfeeding.
  • Do not take if you have liver disease or very high liver enzyme levels in your blood.
  • Tell your doctor if you have kidney problems, as this medicine may further lower blood pressure and hemoglobin levels.

  • If you have a specific lung blood vessel disease (venous occlusive lung disease), this medicine may cause fluid in your lungs (pulmonary edema). Contact your doctor immediately if you experience sudden, severe shortness of breath.
  • Your doctor will arrange for regular blood tests before and during treatment to check for anemia (low red blood cell count) and liver function.
