Leponex is a medicine used to treat schizophrenia when other medicines haven't worked, and to help with severe thought, feeling, and behavior problems in Parkinson's disease.

How to Use

Dosage

Your doctor will start you on a low dose and slowly increase it. For schizophrenia, the usual effective daily dose is 200-450 mg, taken in several doses throughout the day.

Method

Place the orally disintegrating tablet on your tongue and let it dissolve before swallowing, with or without water. Take the tablet immediately after removing it from its packaging.

Important

The maximum daily dose for schizophrenia treatment is 900 mg.

Possible Side Effects

  • Drowsiness
  • Dizziness
  • Increased saliva production
  • Headache
  • Tremors or stiffness
  • Restlessness or difficulty staying still
  • High blood pressure
  • Fainting or feeling weak after changing positions
  • Dry mouth
  • Difficulty urinating or urinary incontinence
  • Fatigue
  • Increased sweating or body temperature
  • Weight gain
  • Blurred vision
RARE
  • Very low white blood cell count (agranulocytosis)
  • Severe constipation that needs medical treatment
  • Stomach pain, swelling, fever, or bloody diarrhea
  • Sudden high fever, stiff muscles, leading to unconsciousness (neuroleptic malignant syndrome)
  • Fast or irregular heartbeat, even at rest, palpitations, breathing difficulties, chest pain, or unexplained tiredness
  • Severe chest pain spreading to arm, jaw, neck, or upper stomach (heart attack symptoms)
  • Nausea, vomiting, loss of appetite, yellow skin/eyes (jaundice), or dark urine (signs of liver problems)
  • Signs of a cold, fever, flu-like symptoms, sore throat, or other infection
  • Seizures or muscle spasms
  • Swelling, especially of the face, mouth, throat, or tongue, which can be itchy or painful (allergic reaction)
  • Difficulty swallowing
  • Sudden drop in blood pressure causing fainting or weakness
  • Persistent, painful erection (priapism)
  • Blood clots in the veins, especially in the legs (swelling, pain, redness), which can travel to the lungs and cause chest pain and breathing difficulties

Important Warnings

  • You are allergic to clozapine or any other ingredient in this medicine.
  • You cannot have regular blood tests to check your white blood cell count.
  • You have previously had a very low white blood cell count (agranulocytosis) caused by medicines (except chemotherapy).
  • You had to stop Leponex treatment before due to severe side effects like very low white blood cell count or heart problems.
  • You are being treated with long-acting antipsychotic injections.

  • You have uncontrolled epilepsy.
  • You have severe heart, kidney, or liver disease.
  • You have paralytic ileus (bowel paralysis and severe constipation).
  • You are taking medicines that affect your bone marrow or reduce your white blood cells.
  • Do not take Leponex if you are unconscious or in a coma.
