How to Use

Dosage

The starting dose is usually 12.5 mg once or twice daily, then gradually increased as directed by your doctor, typically to 200-450 mg daily for schizophrenia or 25-37.5 mg daily for Parkinson's disease.

Method

Swallow the tablet with water.

Important

Maximum daily dose is 900 mg for schizophrenia and 100 mg for Parkinson's disease, as prescribed by your doctor.

RARE
  • Lack of white blood cells (agranulocytosis)
  • Speech disorder (e.g. stuttering)
  • Symptoms of respiratory infection or pneumonia, such as fever, cough, difficulty breathing, wheezing
  • Severe, burning upper abdominal pain from pancreatitis, radiating to the back, with nausea and vomiting
  • Fainting and muscle weakness due to a significant drop in blood pressure (circulatory collapse)
  • Difficulty swallowing (food going down the wrong way)
  • Weight gain or increased obesity
  • Breathing pauses during sleep, possibly with snoring
  • Low red blood cell count (anemia)
  • Restlessness
  • Agitation
  • Confusion
  • Delirium
  • Irregular heartbeat
  • Inflammation of heart muscle (myocarditis) or sac around heart (pericarditis)
  • Fluid buildup in heart sac
  • High blood sugar
  • Diabetes mellitus
  • Blood clot in lungs (thromboembolism)
  • Liver inflammation (hepatitis)
  • Liver disease causing yellow skin/dark urine/itching
  • High blood creatinine kinase
  • Persistent painful erection in men (priapism). Seek medical help if it lasts over 4 hours.
  • Spontaneous bleeding or bruising, which may be a sign of reduced platelet count.
  • Symptoms due to poor blood sugar control (such as nausea, vomiting, stomach pain, extreme thirst, excessive urination, disorientation or confusion).
  • Abdominal pain, cramps, abdominal swelling, vomiting, constipation and difficulty passing gas, which may be signs of bowel obstruction.
  • Loss of appetite, abdominal swelling, abdominal pain, yellow skin, severe weakness and general unwellness. These may be signs of developing liver disease that can lead to sudden liver failure.
  • Nausea, vomiting, tiredness or weight loss, which may be symptoms of kidney inflammation.
  • Increased number of platelets which can lead to blood clots, involuntary movements of mouth/tongue or limbs, obsessive thoughts and repetitive behaviors (symptoms of OCD), skin reactions, swelling in front of the ear (enlarged salivary gland), breathing difficulties, very high levels of triglycerides or cholesterol in the blood, heart muscle disease (cardiomyopathy), cardiac arrest, sudden and unexplained death.
  • Severe chest pain, tightness, pressure, or crushing feeling in the chest (may spread to arm, jaw, neck), shortness of breath, sweating, weakness, confusion, nausea, vomiting, and palpitations (signs of heart attack), which can be fatal. Seek immediate medical attention.

Important Warnings

  • Do not use if you are allergic to clozapine or any other ingredient in Leponex.
  • You cannot have regular blood tests.
  • You have a history of low white blood cell count caused by medicines.
  • You had to stop Leponex before due to serious side effects like very low white blood cells or heart problems.
  • You are taking or have taken long-acting antipsychotic injections.

  • You have or have had a bone marrow disease.
  • You have uncontrolled epilepsy (seizures).
  • You have an acute mental illness from alcohol or drug use.
  • You have reduced consciousness and severe drowsiness.
  • You have circulatory collapse, possibly from severe shock.
  • You have severe kidney disease.
  • You have myocarditis (heart muscle inflammation).
  • You have another severe heart condition.
  • You have symptoms of active liver disease, such as jaundice (yellow skin/eyes), nausea, and loss of appetite.
  • You have another severe liver disease.
  • You have paralytic ileus (bowel paralysis and severe constipation).
  • You are taking medicines that stop your bone marrow from working normally.
  • You are taking medicines that lower your white blood cell count.
  • Contact your doctor immediately if you get signs of chills, fever, flu-like symptoms, sore throat, or other infection symptoms. You will need an urgent blood test.
  • Contact your doctor immediately if you have a sudden rapid rise in body temperature and muscle stiffness, which can lead to unconsciousness (neuroleptic malignant syndrome). This is a serious side effect needing immediate care.
  • Contact your doctor immediately if your heartbeat is fast or irregular, even at rest, or you have palpitations, breathing difficulties, chest pain, or unexplained tiredness. Your doctor needs to check your heart.
  • Contact your doctor immediately if you experience nausea, vomiting, and/or loss of appetite. Your doctor needs to check your liver.
  • Contact your doctor immediately if you have severe constipation. This requires medical treatment to prevent other complications.
  • Contact your doctor immediately if you experience constipation, stomach pain, tenderness in the abdomen, fever, swelling, and/or bloody diarrhea. Your doctor needs to examine you.
