How to Use

Dosage

The usual dose is 1-3 tablets per day. Your doctor may increase the dose up to 6-8 tablets daily, depending on how you respond.

Method

Take the tablets with enough liquid, for example, a glass of water, usually at the same time each day.

The maximum daily dose is 6-8 tablets per day.

  • Feeling depressed, confused, or having behavioral changes or mania
  • Seizures
  • Glaucoma (eye pressure disease) appearing or getting worse
  • Ringing in the ears (tinnitus)
  • Heart rhythm or conduction problems, high blood pressure, dizziness
  • Sour taste in mouth, mouth inflammation, vomiting, nausea
  • Yellow skin or eyes (jaundice), liver inflammation (hepatitis), rash
  • Sensitivity to light
  • Difficulty urinating, urinary incontinence, or changes in sexual desire/ability
RARE
  • Changes in blood cell counts
  • Loss of appetite and weight loss
  • Sweating, high body temperature
  • Memory loss
  • Headache, drowsiness
  • Restlessness, tremor
  • Bowel obstruction
  • Hallucinations
  • Dry eyes

Important Warnings

  • Do not take if you are allergic to amitriptyline, chlordiazepoxide, or any other ingredients in this medicine.
  • Do not take if you have severe muscle weakness (myasthenia gravis), a tumor that produces adrenaline (pheochromocytoma), severe sleep apnea, severe breathing problems, or severe liver problems.
  • Do not take if you recently had a heart attack or have certain heart conditions (like bundle branch block) or untreated glaucoma.
  • Do not take if you are using certain other medicines for depression called MAO inhibitors.
  • Do not use Klotriptyl if you are poisoned by alcohol, sleeping pills, or strong pain relievers (like morphine).

  • Klotriptyl should not be given to children or teenagers under 18 years old.
  • Stop taking this medicine and get medical help right away if you develop a widespread skin rash, fever, and swollen lymph nodes (DRESS syndrome).
  • Tell your doctor if you take other antidepressants or certain pain medicines (opioids), as this combination can lead to a very serious, life-threatening condition called serotonin syndrome.
  • Tell your doctor if you have a slow heartbeat, heart failure, or other heart problems, or if you take medicines that affect your heart, or if you have low potassium or magnesium levels, as Klotriptyl can affect heart rhythm.
  • This medicine can make suicidal thoughts worse, especially when you first start treatment. Contact a doctor right away if this happens.
  • Using this medicine with opioids (strong painkillers) can greatly increase drowsiness, breathing problems, and lead to coma, which can be life-threatening.
  • Klotriptyl can make the effects of alcohol, sedatives, antidepressants, anxiety medicines, antipsychotics, sleeping pills, and antihistamines much stronger, causing more drowsiness.

Use in Elderly Patients

Category D

Avoid use in older adults. Anticholinergic. Sedative. Serotonergic. Risk of QT prolongation. Significant adverse effects include confusion, cognitive impairment, arrhythmias and orthostatic hypotension. Increases risk of falls. Consider the numerous potential interactions.
