Froidir (clozapine) is a medicine used to treat schizophrenia when other treatments haven't worked, and to help with severe thought, emotional, and behavior problems in Parkinson's disease.

How to Use

Dosage

Your doctor will tell you how much to take. For schizophrenia, start with 12.5 mg once or twice a day, gradually increasing to a usual dose of 200-450 mg daily. For Parkinson's, start with 12.5 mg in the evening, gradually increasing to a usual dose of 25-37.5 mg in the evening.

Method

Swallow the tablet whole with water. You can divide the tablet into equal doses if needed.

Important

For schizophrenia, the maximum daily dose is 900 mg. For Parkinson's disease, the maximum daily dose is 100 mg.

Possible Side Effects

  • Severe constipation
  • Fast heartbeat
  • Drowsiness
  • Dizziness
  • Increased saliva
  • Signs of infection like a cold, fever, sore throat, or flu-like symptoms
  • Seizures or convulsions
  • Sudden fainting or loss of consciousness with muscle weakness
  • High white blood cell count
  • Weight gain
  • Blurred vision
  • Headache
  • Tremors, stiffness, restlessness, muscle twitching, abnormal movements, difficulty starting movements, difficulty staying still
  • Changes in heart's electrical activity (ECG)
  • High blood pressure
  • Weakness or unreal feeling after changing position
  • Nausea
  • Vomiting
  • Loss of appetite
  • Dry mouth
  • Slight changes in liver tests
  • Difficulty holding urine or urinating
  • Tiredness
  • Fever
  • Increased sweating
  • Speech problems like slurred speech
RARE
  • Sudden high fever, stiff muscles, or loss of consciousness (Neuroleptic Malignant Syndrome)
  • Strange or unreal feeling, dizziness, or fainting when standing up (increases fall risk)
  • Lack of white blood cells (agranulocytosis)
  • Speech problems like stuttering
  • Signs of lung infection or pneumonia (fever, cough, difficulty breathing, wheezing)
  • Severe upper abdominal pain (pancreatitis)
  • Fainting and muscle weakness from low blood pressure
  • Difficulty swallowing
  • Nausea, vomiting, or loss of appetite (requires liver check)
  • Weight gain or obesity
  • Breathing pauses during sleep (with snoring)
  • Low red blood cell count (anemia)
  • Restlessness, agitation, confusion, delirium
  • Irregular heartbeat, heart muscle or sac inflammation, fluid around the heart
  • High blood sugar (diabetes)
  • Blood clot in the lungs
  • Liver inflammation (hepatitis) causing yellow skin/dark urine/itching
  • High levels of a muscle enzyme (creatine phosphokinase)
  • Persistent painful erection in men (priapism)
  • Increased platelets (risk of blood clots)
  • Spontaneous bleeding or bruising (low platelets)
  • Symptoms from poor blood sugar control (nausea, vomiting, abdominal pain, excessive thirst, frequent urination, confusion)
  • Abdominal pain, cramps, swelling, vomiting, constipation, difficulty passing gas (bowel obstruction)
  • Loss of appetite, abdominal swelling, abdominal pain, yellow skin, severe weakness and discomfort (signs of liver disease, possibly leading to acute liver failure)
  • Nausea, vomiting, tiredness, or weight loss (signs of kidney inflammation)
  • Involuntary movements of mouth/tongue, arms, and legs
  • Obsessive thoughts and compulsive behavior (OCD symptoms)
  • Skin reactions
  • Swelling in front of the ear
  • Difficulty breathing
  • Very high triglyceride or cholesterol levels
  • Heart muscle disease (cardiomyopathy)
  • Cardiac arrest
  • Sudden unexplained death

Important Warnings

  • Do not take Froidir if you are allergic to clozapine or any other ingredients in this medicine.
  • You must have regular blood tests while taking this medicine. Do not take it if you cannot have these tests.
  • Do not take Froidir if you have ever had a low white blood cell count, especially if it was caused by a medicine.
  • Do not take Froidir if you have severe heart, kidney, or liver disease.
  • Tell your doctor immediately if you get signs of infection like a cold, fever, sore throat, or flu-like symptoms, as you may need a blood test.

  • Seek immediate medical help if you have a sudden high fever, stiff muscles, or become unconscious, as this could be a serious side effect.
  • Contact your doctor if your heart beats fast or irregularly, even at rest, or if you have chest pain, breathing problems, or unusual tiredness.
  • Do not use Froidir with other medicines that harm your bone marrow or reduce blood cell production (e.g., carbamazepine, certain antibiotics).
