How to Use

Dosage

The usual starting dose of bortezomib is 1.3 mg per square meter of body surface, given twice a week.

Method

This medicine is given as an injection into a vein or under the skin by a healthcare professional. Injections into a vein are quick, and injections under the skin are given in the thigh or abdomen.

Important

Typically one dose per day on the scheduled treatment days.

Possible Side Effects

COMMON
  • Nerve problems like numbness, tingling, burning, or pain in hands or feet.
  • Feeling tired and weak, sometimes with fever.
  • Nausea (feeling sick), vomiting, or less desire to eat.
  • Stomach problems like constipation (hard to poop, sometimes with bloating) or diarrhea (loose stools). If you have diarrhea, drink plenty of water.
  • Muscle and bone pain.
  • Low red or white blood cell counts, which can lead to tiredness, pale skin, or make you more likely to get infections.
RARE
  • Heart and blood vessel issues: problems with your heart (like heart failure or heart attack), high or low blood pressure, blood clots, bleeding problems, or poor circulation.
  • Kidney and liver issues: changes in how your kidneys or liver work, including swelling or bleeding in the liver, or kidney cancer.
  • Brain and nerve issues: stroke, bleeding in the brain, seizures, paralysis, tremors, confusion, memory loss, severe brain swelling, or nerve damage like Guillain-Barré syndrome.
  • Breathing problems: shortness of breath, wheezing, hiccups, or fast breathing.
  • Allergic reactions: severe allergies with symptoms like trouble breathing, chest pain, dizziness, itching, or swelling of the face, lips, tongue, or throat.
  • Eye and ear problems: irritation, dryness, blurred vision, swelling, bleeding, or loss of vision; hearing loss, ringing in ears, or ear pain/bleeding.
  • Skin and hair changes: rashes, itching, redness, dry skin, hair loss, skin infections, skin cancer, bruises, sores, or abnormal nails.
  • Other serious effects: multi-organ failure, coma, or death.

Important Warnings

  • Do not use this medicine if you are allergic to bortezomib, boron, or any other ingredients, or if you have severe lung or heart problems.
  • This medicine is not for children and teenagers, as its effects on them are not known.
  • Tell your doctor about any past or current health problems, especially low blood cell counts, bleeding issues, stomach problems, dizziness, kidney or liver problems, nerve damage, heart issues, cough, seizures, shingles, tumor lysis syndrome, or memory loss.
  • You will need regular blood tests before and during treatment to check your blood cell counts.
  • If you have had Hepatitis B, tell your doctor, as the infection can return and be serious.

  • Do not use this medicine if you are pregnant or breastfeeding. Men and women must use effective birth control during treatment and for 3 months after.
  • This medicine can make you feel tired, dizzy, or have blurred vision. Do not drive or operate machines if you have these effects, and be careful even if you don't.
