How to Use

Dosage

Adults usually take one 5 mg tablet once a day; your doctor may increase the dose to 10 mg. For children and adolescents 8-18 years old: 5 mg once daily if weighing 35 kg or more, or 2.5 mg once daily if weighing between 20 kg and 35 kg.

Method

Take one tablet whole with water at the same time each day; do not split, crush, or chew it, and you can take it with or without food.

Important

The maximum daily dose for adults is 10 mg; for children, the maximum dose depends on weight, up to 5 mg.

Possible Side Effects

COMMON
  • Headache
  • Dizziness
  • Fast or irregular heartbeat (palpitations)
  • Worsening shortness of breath soon after starting Ambrisentan Accord
  • Runny or stuffy nose, sinus congestion or pain
  • Nausea
  • Diarrhea
  • Tiredness
  • Allergic reactions (rash, itching, swelling of face/lips/tongue/throat, difficulty breathing/swallowing)
  • Swelling (edema), especially of ankles and feet
  • Heart failure (shortness of breath, extreme tiredness, swelling of ankles and legs)
  • Low red blood cell count (anemia) (tiredness and weakness, shortness of breath, general feeling of being unwell)
  • Low blood pressure (dizziness or feeling faint)
  • Blurred vision or other vision changes
  • Fainting
  • Abnormal liver test results
  • Constipation
  • Stomach pain
  • Chest pain or discomfort
  • Flushing (reddening of skin)
  • Vomiting
  • Feeling of weakness
  • Nosebleeds
  • Rash
  • Ringing in the ears (tinnitus) (when used with tadalafil)
RARE
  • Liver damage
  • Inflammation of the liver caused by the body's own immune system (autoimmune hepatitis)
  • Sudden hearing loss (when used with tadalafil)

Important Warnings

  • Do not use if you are allergic to ambrisentan, soy, or any other ingredients in the medicine.
  • Do not use if you are pregnant, planning to become pregnant, or breastfeeding, and use reliable contraception if you can become pregnant.
  • Talk to your doctor if you have liver problems or liver disease.
  • Talk to your doctor if you have anemia (low red blood cells in your blood).
  • Tell your doctor if you have swelling in your hands, ankles, or feet due to fluid retention.

  • Talk to your doctor if you have certain lung conditions, such as pulmonary veno-occlusive disease or idiopathic pulmonary fibrosis.
  • This medicine should not be given to children under 8 years of age.
  • You will need regular blood tests to check your liver function and blood during treatment.
  • If you notice signs of liver problems, such as loss of appetite, nausea, yellow skin/eyes, or dark urine, contact your doctor immediately.
  • Do not drive or operate machinery if you feel unwell, dizzy, or tired.
