How to Use

Dosage

Adults typically take one 5 mg tablet once a day. Your doctor might increase this to 10 mg once a day. For children (8-18 years), the dose depends on weight: 5 mg daily for those weighing 35 kg or more, and 2.5 mg daily for those weighing 20 kg to under 35 kg.

Method

Take the tablet whole with water at the same time each day. Do not split, crush, or chew it. You can take it with or without food.

Important

The maximum daily dose for adults is 10 mg. For children, the maximum dose depends on their weight and medical condition, but typically does not exceed 5 mg.

Possible Side Effects

COMMON
  • Headache
  • Dizziness
  • Swelling, especially of the ankles and feet
  • Allergic reactions (rash, itching, swelling of face/lips/tongue/throat, difficulty breathing/swallowing)
  • Heart failure (shortness of breath, extreme tiredness)
  • Low red blood cell count (anemia, symptoms include tiredness, weakness, shortness of breath, general malaise)
  • Low blood pressure (dizziness, feeling faint)
  • Palpitations (fast or irregular heartbeat)
  • Worsening shortness of breath soon after starting treatment
  • Runny or blocked nose, sinus congestion or pain
  • Nausea
  • Diarrhea
  • Tiredness/Fatigue
  • Flushing (skin redness)
  • Vomiting
  • Chest pain or discomfort
  • Blurred vision or other vision changes
  • Fainting
  • Abnormal liver test results in blood tests
  • Constipation
  • Abdominal pain
  • Feeling of weakness
  • Nosebleed
  • Rash
  • Ringing in the ears (tinnitus)
RARE
  • Liver damage
  • Inflammation of the liver caused by the body's own immune system (autoimmune hepatitis)
  • Sudden hearing loss

Important Warnings

  • Do not take if you are allergic to ambrisentan, soy, or any other ingredient in this medicine.
  • Do not take if you are pregnant, planning to become pregnant, or could become pregnant without reliable contraception, as it can harm the unborn child.
  • Do not breastfeed while taking this medicine.
  • Do not take if you have severe liver disease. Discuss any liver problems with your doctor.
  • Do not take if you have lung scarring of unknown cause (idiopathic pulmonary fibrosis).

  • You will need regular blood tests to check for anemia and liver function. Contact your doctor immediately if you notice signs of liver problems (e.g., loss of appetite, nausea, yellow skin/eyes, dark urine, itchy skin).
  • Do not give this medicine to children under 8 years of age.
  • This medicine can cause low blood pressure, dizziness, and tiredness, which may affect your ability to drive or operate machinery. Do not drive or use machines if you do not feel well.
